Also made Primary Cell Culture this week:

  • For these cells, the egg laying scenario was slightly different:
    • Flies were put on plates in mid afternoon Saturday and left overnight
    • Then flies were swapped to a new plate in the morning on Sunday and left for 24 hours
    • Plates were not collected until at least 10:30am to facilitate morning laying
    • Then plates were left to “mature” (eggs to cellularize) for 2-3 hours after being taken off before the primary cell culture process began
    • The second cc plate was put on regularly
  • On 05/09 2 primary cc flasks were make for SPZ flies, and 2 for innubila
  • On 05/10 3 primary cc flasks were make for SPZ flies, and 2 for innubila

  • Looking good, places with nice alive growing cells

  • Also looking good and there are lots of places where cells seem to still be growing

04/04 secondary from 01/27

  • 2 flasks
  • Basically nothing, barely any cells and lots of precipitate
  • I did not do a good job of scraping up the primary flask
  • Flasks were thrown out

02/24 primary cell scraped

  • 1 flask
  • Flask looks a little strange but cells are still growing

02/25 primary cell scraped

  • 1 flask
  • Flask is very sparce but there are some clumps of cells alive/growing

  • Cells are alive and sort of growing but at low density

  • No cells attached to the flasks

  • Flask is sparce and there are a lot of floating clumps
  • But there is at least 1 clump contracting (see pic)

05/03 primary

  • 3 flasks
  • Lots of clumps that look alive but aren’t sticking to the flask!
  • There is a clump contracting though (see pic)

05/09 and 05/10 primary

  • 2 flasks each
  • Very early to tell
  • Some alive clumps of cells
  • Some huge debris and dead cell clumps

  • Cells look alive in some places but are kind of small

  • 1 flask
  • Cells still alive in here, but little or very slow growth!

  • Clump contracting in these flasks
  • Not much is growing, cells are sparce and there are a lot of floating clumps

05/03 domeless primary

  • 3 flasks
  • Everything is dead and shriveled in these flasks

  • No real growth yet but cell clumps look to be alive, cells are pretty small
